Johnson, Terry C. (University of Minnesota, Minneapolis), and Leroy C. McLaren. Plaque development and induction of interferon synthesis by RMC poliovirus. J. Bacteriol. 90:565-570. 1965.-Plaque development by RMC poliovirus on human amnion cell monolayers was investigated with regard to autointerference and to the effect of acid-agar overlay on plaquing efficiency. The virus was inhibited by acid-agar overlay, thereby exhibiting the d(-) marker typical of attenuated poliovirus strains. In addition, a lack of RMC poliovirus plaque development on HeLa cell monolayers was shown to be the result of an agar inhibitor which could be removed by NaCl extraction. By use of a simplified plaque reduction assay, it was shown that interferon production was responsible for the autointerference phenomenon. Interferon synthesis did not correlate with the ages in vitro of human amnion cell cultures. Fibroblasts originating from the chorionic membrane produced negligible amounts of the inhibitor. Interferon synthesis by human amnion cells infected with RMC poliovirus was inhibited by actinomycin D. The addition of guanidine hydrochloride to infected cultures immediately after RMC poliovirus adsorption markedly inhibited interferon synthesis, although after 2 hr (postadsorption) guanidine had no effect on interferon production.

Poliovirus Particles do not Form in Preinfected Cells with Reovirus
Background and Aims: Inhibition of viral growth in coinfected cells with two different viruses has been described. This phenomenon known as viral interference can occur in several virus host systems such as interference of enterovirus infection on poliovirus vaccine strains. In this study we superinfected reovirus infected HeLa cells with poliovirus to determine if poliovirus can replicate in s...
متن کاملMechanism of enhanced plaque formation by poliovirus in poxvirus-infected cells.
The mechanism of the enhancement by Yaba virus (YV) of poliovirus plaque formation was studied. Poliovirus adsorbed equally well to both YV-infected and normal J INET cells. Infectious RNA of poliovirus was also enhanced in YVinfected cells, thus indicating the step or sequence of steps of poliovirus growth cycle enhanced by YV may be those subsequent to poliovirus uncoating. One-step growth ex...
متن کاملAssay of human interferon in Vero cells by several methods.
Four methods for the assay of human interferon in Vero cells were compared based on the inhibition of viral cytopathic effect (CPE) in tubes, the inhibition of CPE in microplates, the reduction of plaques, and the inhibition of quantitative hemadsorption. For inhibition of CPE, Sindbis virus, vesicular stomatitis virus, poliovirus type 2, and vaccinia virus were used for challenge. In the plaqu...

متن کاملSequential suppression by actinomycin D of interferon production and cellular resistance induced by Poly I:C.
Cellular RNA and protein synthesis are required both for interferon synthesis (Heller, 1963; Wagner, 1963) and the expression of the antivirus activity of interferon (Taylor, 1964). The suppression of the antivirus activity of interferon by metabolic inhibitors has been interpreted to mean that interferon acts as a de-repressor for the synthesis of a cellular antivirus protein. While direct pro...
